We propose to test a hypothesis following from our preliminary results that the common PCR in vitro simulates the pathological microsatellite expansions in the human genome. We will use PCr and human genome bioinformatics for purpose. Perform PCR in vitr o of about 1000 DNA fragments 10-60 nucleotides in lenght occuring pathologically expanding human genome regions. Generalize the results into rules precting propensity to the expansion to the nucleotide sequence. Carry out a bioinformatic analysis of the human genome. Find whether the microsatellite frequency of occurrence and lenght correlates with the expansion during PCR in vitro. Generalize the results of the project into rules assigning propensity to expansion to the fragments in the human genome. Analyze selected DNA fragments by biophysical methods to understand the mechanism of expansion. |
