The aim of this research project is the adoption of molecular genetics technology in the preparation of mutant animals by the targeted manipulation of the mammalian genome (so-called "gene targeting"). Simultaneously, methodical procedures are developed for optimization of transgenic animals production by microinjection of egg nucleus, microinjection of sperm, transfer of embryonic stem cell into blastocyst, and conjugation of DNA with sperms. The project is also focused on improving cryo-conservation techniques for maintaining the gene pool bank. Another component of the project is preparation, testing and development of recombinant plasmid vectors and phage vectors intended for the construction of target-mutation animals, and development of modified recombinant vectors and their products as models for optimization for protein purification from transgenic animals. |
